An improved cytogenetic technique intended for macroalgal chromosomes

Abstract
The selection of fixative was exercised Therefore, Different fixatives such as acetic acid, acetone, chloroform, methanol, ethanol and formaldehyde were used separately or in combination in altered ratios to optimize it for chromosome staining in macroalgae (seaweeds). Combination of acetic acid with formaldehyde (1:1) was found to be effective for chromosomes staining in Chlorophyta and Rhodophyta. Moreover, the corbol fuchsins and aceto-iron–haematoxylin-chloral-hydrate were found to be effective stains for Rhodophyta and Chlorophyta respectively. These techniques were used to prepare karyological studies in seaweeds including seven Ulva spp., Gracilaria spp., Monostroma spp. and a few Porphyra species.
